Please, if a reserve a slot and than (if it is setted up) and I receive a confirmation request by email, coud the slot be “not available” only after my confirmation affermative answer?

I think you can use default reservation state to be unconfirmed. This slot will be available for booking until admin changes is to ‘confirmed’ or user confirms by emailed link. Will test that lates as I do not use this exact scenario currently.
